چکیده

This article investigates various multiple testing procedures for the testing of unity odds ratios from stratified studies. These procedures compare individual ordered p-values to a series of adjusted nominal levels, and are employed to distinguish between significance and insignificance in stratified studies. Three commonly used approaches for determining p-values are considered: the asymptotic, exact, and mid-P approaches. We focus on two-sided hypothesis testing problems. The empirical performance of various multiple testing procedures is evaluated in terms of their empirical sizes and powers. As expected, multiple testing procedures based on the exact p-value approach are always conservative in the sense that their empirical sizes are always less than or equal to the pre-assigned nominal levels. Interestingly, in some cases we observe that some procedures based on the asymptotic p-value approach are more conservative and less powerful than the associated procedures based on the exact p-value approach. In addition, some procedures based on the mid-P approach are recommended because their empirical sizes are generally close to the pre-assigned nominal level but are the most powerful. General guidelines are provided, and numerical examples are presented to illustrate the methodologies. © 2005 Elsevier B.V. All rights reserved.

Sample size and optimal designs in stratified comparative trials to establish the equivalence of treatment effects among two ethnic groups.

When a new investigational medicine is intended to be applied to populations with different ethnic backgrounds, a stratified comparative phase III trial using ethnic groups as strata may be conducted to assess the influence of ethnic factors on clinical outcomes of this new medicine. In this paper, based on a binomial model with odds ratio as the measure of the treatment effect, we derive the s...

Overview of the validation procedures for a vaccine production: from R&D level to the pre-qualification stage

Just like any other process, vaccine manufacturing procedures are defined as a series of interrelated functions and activities using a variety of specified actions and equipment designed to produce a defined product. To assure the reproducibility and consistency of such processes, they must be carried out using validated equipment and under the established procedures that meet all the acceptanc...

Tenecteplase versus reteplase in acute myocardial infarction: A network meta-analysis of randomized clinical trials

Background: Acute myocardial infarction (AMI) is the leading cause of death throughout the world. One of the standard approaches to treatment of AMI is fibrinolysis. The study was conducted to evaluate the clinical efficacy of tenecteplase versus reteplase through network meta-analysis for AMI. Methods: Randomized trials were comprehensively searched in PubMed, Scopus, Cochrane library, a...

Improving efficiency of inferences in randomized clinical trials using auxiliary covariates.

The primary goal of a randomized clinical trial is to make comparisons among two or more treatments. For example, in a two-arm trial with continuous response, the focus may be on the difference in treatment means; with more than two treatments, the comparison may be based on pairwise differences. With binary outcomes, pairwise odds ratios or log odds ratios may be used. In general, comparisons ...

Properties of the urn randomization in clinical trials.

In this article we review the important statistical properties of the urn randomization (design) for assigning patients to treatment groups in a clinical trial. The urn design is the most widely studied member of the family of adaptive biased-coin designs. Such designs are a compromise between designs that yield perfect balance in treatment assignments and complete randomization which eliminate...
